Enhancing Effect of Nicotinic Acid on the Reduction of Methemoglobin by Rabbit Erythrocytes in viro

Abstract

|
The reduction of methemoglobin by erythrocytes was studied by incubating rabbit erythrocytes at 37¡ÆC for 4 hours in thepresence of a purified human methemoglobin, glucose and methylene blue. At intervals during incubation, aliquots of incubating mixture were removed to determine methemoglabin concentration.
The rate of methemoglobin reduction was markedly increased when the erythrocytes were pre-incubated for 4 hours in reaction media containing glutamine and phosphate buffer (pH 7.4). Nicotinic acid added to the preincubating media enhanced further this process.
These results were interpreted as indicating that NAD in erythrocytes plays an important role in reducing methemoglobin and maintaining normal ferrohemoglobin since nicotinic acid increases primarily the synthesis of NAD in erythrocytes.
